Periodic acid-schiff staining is a common histochemical staining method, which is mainly used to prove and identify the character of intracellular vacuoles, the diagnosis of cardiomyopathy and other cardiovascular diseases, the diagnosis and research of glycogen accumulation disease and the research of diabetes as well as diagnosis of certain tumors, and so on.
The periodic acid-schiff staining liquid is mainly used to show glycogen and other polysaccharides. The periodic acid oxidizes the polysaccharide glycol base group in the cell to an aldehyde group. The aldehyde group is combined with the colorless magenta of Shiff’s solution, stained red and localized on the cytolymph.
a)Short staining time, fast staining, bright stain color
b)The nucleus is bright blue without differentiation
c)Hematoxylin is not affected by storage temperature. After low temperature or freezing, there is no sediment and does not impact the color of staining.
