CK5/6 is mainly expressed in the cell membrane and cytoplasm of squamous epithelial cells or squamous cells. It is almost not expressed in glandular epithelium. CK5/6 has a good differential diagnosis effect for squamous cell carcinoma and adenocarcinoma.

p40 (ΔNp63) is a subtype of p63 protein, which is mainly expressed in bronchial mucosa basal cells of normal lung tissue and glandular myoepithelial cells. It has high specificity and sensitivity to lung squamous cell carcinoma. The combination of p40 (ΔNp63) and CK5/6 can achieve higher sensitivity and specificity.

When used with TTF-1 and Napsin A, CK5/6/p40 antibody reagent can help differentiate poorly differentiated lung adenocarcinoma and squamous cell carcinoma.
